Delft – September 15, 2026 -- AdJane Biosciences has published a White Paper outlining a strategy to add a proprietary intranasal booster to existing intramuscular respiratory vaccines, targeting the mucosal immunity gap left by current mRNA shots.

Phase I data show intranasal booster induced significant nasal IgA responses in mRNA-primed subjects

In a post-hoc subset of participants from AdJane's completed Phase I study who had previously received Pfizer and/or Moderna COVID-19 mRNA vaccines, a subsequent intranasal dose of an Nm-nOMV vaccine displaying SARS-CoV-2 Spike was well tolerated and generated significant nasal IgA responses. AdJane says this provides initial human evidence for a heterologous prime-boost approach: systemic priming via existing intramuscular vaccines followed by a mucosal Nm-nOMV booster delivered intranasally.

Company argues intramuscular vaccines protect against severe disease but leave mucosal surfaces exposed

Current intramuscular respiratory vaccines generate strong systemic immunity but generally induce limited immunity at the mucosal surfaces where respiratory pathogens first enter the body. AdJane cites growing evidence that mucosal IgA responses can reduce transmission at the point of entry, an outcome public health bodies have prioritized alongside reducing severe disease.

Strategy preserves existing vaccine infrastructure while adding a proprietary mucosal layer

Rather than replacing established respiratory vaccine franchises, AdJane's approach retains their infrastructure and broad coverage while layering on a booster designed to induce complementary mucosal immunity. The company says this could open a development path for next-generation vaccination regimens built on proven systemic vaccines.

AdJane has filed patents covering combinations of OMV technology with mRNA and respiratory vaccines

The company holds a broad intellectual property portfolio around its Nm-nOMV platform, covering the platform itself, manufacturing, mucosal dosing and specific vaccine applications. Separate patent filings specifically address combinations of OMV technology with mRNA vaccines and respiratory vaccines.

Potential applications could extend beyond COVID-19 to influenza, RSV and combination vaccines

While the initial clinical observations involve individuals primed with COVID-19 mRNA vaccines, AdJane notes limited mucosal immunity is a feature of many intramuscular respiratory vaccination strategies, not unique to mRNA platforms. Subject to further development and clinical validation, the company says applications could extend across COVID-19, influenza, RSV, combination respiratory vaccines and pandemic preparedness programs.
