SAN FRANCISCO – November 03, 2026 -- Thryv Therapeutics Inc. has raised a $30 million Series B financing led by InClin, Inc., a global contract research organization, to move its SGK1 inhibitor THRV-1268 into a Phase 2a heart failure study that opens for enrollment in early 2027.
The round follows completion of enrollment in the first efficacy cohort of the Wave II Phase 2/3 study of THRV-1268 in Long QT Syndrome. It also follows a favorable review of available Wave II data by the study's Independent Data Monitoring Committee (IDMC).
InClin brings clinical operations alongside capital
InClin, which operates in San Mateo, California and Sydney, Australia, joins as a strategic investor. In recent months Thryv has worked with the CRO to establish clinical operations in Australia, enabling initial patient enrollment in ASPIRE-HF early next year.
Proceeds will fund the study through a pre-specified analysis of safety and efficacy, including changes in QTc and established biomarkers of heart failure risk. Existing investors Fonds de solidarité FTQ, Amplitude Ventures, Investissement Québec, Lumira Ventures, adMare BioInnovations, CTI Life Sciences Fund and Alexandria Venture Investments continue to back the company.
ASPIRE-HF randomizes patients to two THRV-1268 doses or placebo over 24 weeks
ASPIRE-HF is a double-blind, randomized, placebo-controlled Phase 2a study. It enrolls adults with symptomatic heart failure with reduced ejection fraction (HFrEF) and QTc prolongation who are on guideline-directed medical therapy (GDMT). The study will assess safety, pharmacokinetics, and effects on markers of arrhythmic risk, cardiac function and cardiac remodeling. A pre-specified early efficacy evaluation is set for Week 6.
Chief Medical Officer Amy Sehnert called ASPIRE-HF "the culmination of years of work to establish SGK1 as a therapeutic target." She cited human genetic and preclinical evidence, plus Phase 1 results showing QT interval shortening in obese individuals, as support for expanding into heart failure.
Wave II completes first efficacy cohort enrollment, with data due in early 2027
The Wave II Phase 2/3 study in Long QT Syndrome (NCT07277582) has finished enrolling its first efficacy cohort. In the third quarter of 2026, the IDMC reviewed available safety and pharmacokinetic data and recommended continuing without modification. No serious adverse events have been reported.
Thryv has submitted a protocol amendment to its U.S. FDA IND to include patients with Long QT Syndrome Type 3. The study is now open to participants aged 15 years and older.
Thryv takes SGK1 data to AHA Scientific Sessions in Chicago
Thryv will present at the American Heart Association Scientific Sessions 2026, held November 6-9 at McCormick Place Convention Center in Chicago. A late-breaking basic science session will cover SGK1 inhibition, as monotherapy or combined with incretin agonism, to target residual risk in cardiometabolic heart failure. A poster will present initial findings from the myQTwave National Survey on the lived experience and symptom burden of patients with Long QT Syndrome Type 2 and Type 3.
