New York and Suzhou, China – September 20, 2026 -- Accro Bioscience Inc. has dosed its first patient in a Phase IIb trial testing AC-101, a selective RIPK2 inhibitor, in patients with moderate-to-severe ulcerative colitis (UC).
Trial spans roughly 50 clinical sites with three treatment arms
The multicenter, double-blind, placebo-controlled study is designed across approximately 50 clinical sites and includes two AC-101 dose cohorts plus a placebo control arm. The primary endpoint is clinical remission rate at Week 12, with exploratory endpoints covering histologic remission and long-term maintenance of clinical and histologic remission.
AC-101 targets NOD/RIPK2 signaling implicated in inflammatory bowel disease
AC-101 emerged from Accro Bioscience's proprietary drug discovery platform focused on regulatory cell death and inflammation. Receptor interacting protein kinase 2 (RIPK2) is a key mediator in the NOD signaling pathway, and dysregulated NOD/RIPK2 signaling has been linked to multiple inflammatory and autoimmune diseases, including inflammatory bowel disease.
Prior Phase I and Phase Ib data set the stage for the current study
AC-101 has completed Phase I studies in healthy participants in Australia and China, reporting a favorable safety and PK/PD profile. A Phase Ib proof-of-concept study in Chinese patients with moderate-to-severe UC has also been completed, with efficacy and safety results scheduled for presentation at UEG Week 2026.
